Abstract
INTISARI Prayoga, Julian Dhimas, 2023. Perbandingan Nilai Agregasi Trombosit Pada Tikus Galur Wistar (Rattus norvegicus) dengan Diet Standar dan Tinggi Lemak. Program Studi D3 Analis Kesehatan. Fakultas Ilmu Kesehatan. Universitas Setia Budi. Hiperkolesterolemia bertanggung jawab terhadap sepertiga kasus stroke iskemik dan diperkirakan menyebabkan 2,6 juta kematian di dunia. Peningkatan kadar kolesterol juga menyumbang 29,7 juta (2%) dari jumlah kasus global. Hiperkolesterolemia dapat menyebabkan disfungsi endotel yang dapat memicu terjadinya peningkatan nilai agregasi trombosit yang melakukan peran penting dalam terjadinya penyakit kardiovaskuler. Penelitian ini bertujuan untuk mengetahui perbedaan nilai agregasi trombosit antara tikus galur wistar (Rattus norvegicus) yang diberi diet standar dan diet tinggi lemak. Penelitian dilaksanakan pada bulan Februari sampai dengan April 2023 di Laboratorium Universitas Setia Budi dilakukan dengan menggunakan metode SADT dengan perhitungan rumus velaskar. Populasi yang digunakan adalah tikus galur wistar dengan kriteria jantan dengan berat ≥ 200 gram sebanyak 10 tikus yang dibagi menjadi 2 kelompok yaitu kelompok dengan diet standar dan tinggi lemak. Kelompok tikus standar diberikan diet dengan pakan standar sedangkan kelompok tikus dengan diet tinggi lemak diberi pakan standar dengan penambahan lemak babi dan kuning telur bebek selama 4 minggu. Selanjutnya dilakukan pemeriksaan nilai agregasi trombosit pada kedua kelompok lalu dilakukan analisis data secara statistik dengan uji beda independent sample t-test. Hasil analisis independent sample t-test didapatkan nilai p = 0,002 sehingga dapat disimpulkan terdapat perbedaan signifikan nilai agregasi trombosit pada kelompok tikus dengan diet standar dan tinggi lemak. Hypercholesterolemia is responsible for one third of ischemic stroke cases and is estimated to cause 2.6 million deaths worldwide. Elevated cholesterol levels also accounted for 29.7 million (2%) of the total global cases. Hypercholesterolemia can cause endothelial dysfunction which can trigger an increase in platelet aggregation values which play an important role in the occurrence of cardiovascular disease. This study aims to determine differences in platelet aggregation values between wistar strain rats (Rattus norvegicus) fed a standard diet and a high-fat diet. The research was carried out from February to April 2023 in Setia Budi University Laboratory was using the blood smear method with the calculation of the velaskar formula. The population used was Wistar rats with a mixture of males weighing ≥ 200 grams as many as 10 rats which were divided into 2 groups, namely the group with a standard diet and high fat. The group of rats was given a standard ration with standard feed while the group of rats with a high-fat ration was given a standard diet with the addition of lard and duck egg yolk for 4 weeks. Furthermore, the value of platelet aggregation was examined in both groups and then the data were analyzed statistically using the independent sample t-test. The results of the analysis of the independent sample t-test obtained the p = 0.002 therefore it can be concluded that there is a significant difference in the value of platelet aggregation in the rat group with a standard and high-fat diet Keywords: platelet aggregation, wistar rats, high-fat diet vii
