Abstract
xvi ABSTRACT Jentewo, CM. 2023. Differences in the Histopathological Picture of Chicken Liver with Time Variations of 10% Formalin Neutal Buffer Fixation in Hematoxylin- Eosin Staining. Bachelor of Applied Science Medical Laboratory Technology, Faculty of Health Sciences, Setia Budi University. Histopathological examination is a gold standard to determine patient diagnosis, especially for cancer diagnosis with the technique used, namely histotechnics. One of the most important stages in histotechnics is fixation because fixation aims to keep tissue decay from occurring. The ideal time used to fix is for 8 to 24 hours. Fixation done less than normal time causes the tissue not to be fixed properly and if it exceeds normal time it will result in over fixation which will affect the coloring result. The purpose of this thesis research is to determine the difference between the histopathic picture of chicken liver with the time variation of 10% Neutral Buffer Formalin fixation in Hematoxin-Eosin staining. This research was conducted at the Anatomical Pathology Laboratory of Dr Soeratno Gemolong Hospital, using three treatments with a descriptive analysis of the microscopic picture of chicken liver histopathology with Hemactosilin-Eosin staining. Treatment I is fixed for 4 hours. Treatment II as a control is fixed for 24 hours. And Treatment III is fixed for 96 hours (4 days). Based on the results of research that has been obtained, it shows that fixation time affects the microscopic picture of tissues. Fixation with a length of 24 hours gives a good microscopic picture, while for fixation time 4 hours and 96 hours gives a microscopic picture that is not good because it occurs under fixation and over fixation. So the length of time fixation is very influential on the microscopic results of tissues.
