D4 Study Program of Study Health Analyst, Faculty of Health Sciences, Setia Budi University, Surakarta. Fixation is a complex series of chemical processes, bringing about changes in the various chemical constituents of the cell, but in the morphology of the cell. NBF solution is a commonly used mixed fixation agent that functions as a preservative and to protect the physical structure of cells. Bouin's solution is considered a good fixative for delicate soft tissue structures. The purpose of this study was to determine the difference between the use of bouin fixation fluid and buffered formalin on the histological appearance of mice kidneys (mus musculus) stained with hematoxylin eosin. This type of research is included in experimental research and its criteria, such as descriptive research and sampling techniques. In this study, the kidney tissue of mice (mus musculus) first carried out the process of making the tissue then carried out the fixation process with bouin solution for 6 hours and 10 hours while buffered formalin for 36 hours and 48 hours. The variable used in this study was the independent variable in this study, fixation of buffered formalin solution and Bouin solution, and the dependent variable was the histological appearance of the mice kidney (mus musculus). The results of this study in fixation with bouin solution, the nucleus and cytoplasm were less clear and there were artifacts, while in formalin buffered fixation, the nucleus and cytoplasm were clear but pale. The conclusion of this study was that there were differences in the microscopic appearance of the kidney tissue preparations of mice (mus musculus) which had been fixed with neutral buffered formalin showing clear images of the cell nucleus and cytoplasm, whereas in the microscopic images fixed with bouin it was less clear and there were visible artifacts.
