Abstract

Nurprabowo, R. 2023. Effect of Sputum Sample Storage Time for Tuberculosis Examination with the ZN Method (Ziehl Neelsen). D4 Health Analyst Study Program, Faculty of Health Sciences, Setia Budi University. Tuberculosis is a direct infectious disease caused by TB bacteria, namely Myobacterium tuberculosis. The background of the researchers conducting research at the UPTD Health Laboratory in the Magetan Regency area was due to the frequent or large number of examinations being stored. The aim of the study was to determine the effect of storage time for sputum samples for tuberculosis examination using the ZN (Zihl-Neelsen) method microscopically which was stored for 8 hours room temperature and 8 hours at 2oC – 8oC at UPT Regional Health Laboratory of Magetan Regency. Research data was in the form of primary data in the form of sputum samples from the UPTD Health Centers in Magetan district which were sent to the UPT Regional Health Laboratory in Magetan Regency. The effect of storage time for sputum samples for tuberculosis examination using the Zihl Neelsen method is interpreted using the International Union Against Tuberculosis and Lung Disease (IUATLD) international scale. The results of the 30 samples that were tested for TCM showed that 22 samples were confirmed positive and ZN was then examined with 3 treatments. Storage 0 hours to 8 hours at room temperature there is a change in the interpretation of the results where the number of bacteria increases causing a change in the positive results, whereas from storage 0 hours to 8 hours at 2 oC – 8 oC the results are exactly the same as the interpretation of the results, there are even samples in which the number of bacteria decreased and increased, but did not change the interpretation of a positive result in the sample. Based on the results of the three treatments above, it can be concluded that there was no effect on the storage time of the sputum samples used for tuberculosis examination, but with the recommendation that samples should be stored at a temperature of 2 oC – 8 oC..
