Abstract

CLAUDYA M. W., 2023. UJI TOKSISITAS AKUT EKSTRAK ETANOL KULIT SALAK PONDOH (Salacca zalacca.) TERHADAP TIKUS PUTIH GALUR WISTAR, SKRIPSI, FAKULTAS FARMASI, UNIVERSITAS SETIA BUDI, SURAKARTA. Kulit buah salak berpotensi sebagai obat tradisional karena senyawa yang dikandung didalamnya berupa senyawa-senyawa yang memiliki banyak manfaat yaitu sebagai antimikroba, memiliki aktivitas antioksidan, dan berpotensi dapat menurunkan kadar gula darah dan menurunkan kolestrol. Tujuan dari penelitian ini adalah untuk mengetahui efek toksik, dan nilai LD50 dari ekstrak etanol kulit salak pondoh terhadap tikus putih jantan. Penelitian ini menggunakan metode maserasi. Kulit salak dimaserasi dengan pelarut etanol 70% hingga diperoleh ekstrak kental. Kemudian ekstrak dilakukan pengujian senyawa kimia mengunakan metode KLT (Kromatografi Lapis Tipis). Pada uji toksisitas akut dilakukan dengan metode fixed dose. Ekstrak kulit salak dosis 5, 50, 300, 2000 mg/kgBB diberikan pada tikus jantan. Jumlah hewan uji yang digunakan adalah 25 ekor tikus, jenis kelamin jantan dan dibagi menjadi 5 kelompok. Pengamatan dilakukan selama 24 jam dan dilanjutkan hingga 14 hari dengan mengamati gelagat yang ditimbulkan pada hewan uji, pengamatan berat badan, kematian hewan uji, dan perubahan makropatologi pada organ tikus putih jantan. Hasil pengamatan uji toksisitas akut ekstrak kulit salak pondoh, ditemukan adanya gelagat, penurunan berat badan, dan perubahan warna organ, serta kematian pada tikus putih jantan. Kesimpulan dari hasil penelitian adalah ekstrak kulit salak menimbulkan efek toksik pada tikus putih jantan. CLAUDYA M. W., 2023. Salak fruit peel has the potential as a traditional medicine because the compounds contained in it are compounds that can cure various diseases. The purpose of this study was to determine the toxic effect, and the LD50 value of the ethanol extract of pondoh bark on white rats. This study used the maceration method. Salak peels were macerated with 70% ethanol fuel to obtain a thick extract. Then the extract was tested for chemical compounds using the TLC (Thin Layer Chromatography) method. The acute toxicity test was carried out using the fixed dose method. Salak bark extract doses of 5, 50, 300, 2000 mg/kg BW were given to male rats. The number of test animals used was 25 rats, male sex and divided into 5 groups. Observations were made for 24 hours and continued for up to 14 days by observing the signs evoked in animal tests, body weight observations, animal death tests, and macropathological changes in the organs of male white rats. The results of observations of the acute toxicity test of pondoh bark extract, found signs, weight loss, organ discoloration, and death in male rats. The conclusion from the research results is that the bark extract of salak has a toxic effect on male white rats.
