Abstract

NOVA FATIHA RAHMADANI, 2025, FORMULASI SEDIAAN GEL NATRIUM DIKLOFENAK DENGAN VARIASI KONSENTRASI Gelling Agent HPMC TERHADAP MUTU FISIK SEDIAAN, KARYA TULIS ILMIAH, PROGRAM STUDI D-III FARMASI, FAKULTAS FARMASI, UNIVERSITAS SETIA BUDI SURAKARTA. Dibimbing oleh Dr. apt. Ilham Kuncahyo, S.Si., M.Sc. Natrium diklofenak merupakan golongan NSAID (Non Steroidal Anti Inflamatorry Drugs) atau AINS (Anti Inflammatory Non Steroid) yang memiliki mekanisme kerja sebagai antiinflamasi, analgetik dan antipiretik. Penggunaan natrium diklofenak secara oral memiliki efek samping mengiritasi lambung dan waktu paruh yang pendek. Maka untuk memecahkan suatu masalah dibuat sediaan topikal dengan konsentrasi 1% dari natrium diklofenak untuk menghindari efek samping tersebut. Penelitian ini menggunakan HPMC sebagai gelling agent dengan variasi konsentrasi untuk menghasilkan mutu fisik gel yang baik. Gel natrium diklofenak dibuat 3 formula dengan variasi konsentrasi dari HPMC antara lain 1,5%, 2,5%, dan 3,5% dengan dilakukan pengujian mutu fisik meliputi uji organoleptis, uji homogenitas, uji pH, uji viskositas, uji daya lekat, uji daya sebar, dan uji stabilitas real time. data tersebut akan dilakukan analisis menggunakan sistem SPSS dengan ANOVA one way dan uji Paired T-Test. Hasil penelitian menunjukkan bahwa variasi konsentrasi HPMC sebagai gelling agent dapat mempengaruhi kualitas mutu fisik sediaan gel yang mencakup viskositas, daya sebar, daya lekat, dan pH, akan tetapi tidak memengaruhi terhadap organoleptis dan homogenitas. Semakin tinggi konsentrasi HPMC yang diterapkan, viskositas, daya lekat, dan pHnya juga akan meningkat. Namun daya sebar gel menjadi rendah karena berbanding terbalik dengan viskositas. Hasil penelitian memberikan formula yang baik pada formula 1 (HPMC 1,5%) dan dan formula 2 (HPMC 2,5%). Supervised by Dr. apt. Ilham Kuncahyo, S.Si., M.Sc. Diclofenac sodium belonged to a class of NSAIDs (Non Steroidal Anti Inflammatory Drugs) or AINS (Anti Inflammatory Non Steroid) which had a mechanism of action as anti-inflammatory, analgesic, and antipyretic. Oral use of diclofenac sodium had side effects of irritating the stomach and a short half-life. To solve the problem, a topical preparation with a concentration of 1% of diclofenac sodium was made to avoid those side effects. This study utilized HPMC as a gelling agent with various concentrations to produce good physical quality of the gel. Diclofenac sodium gel was made with 3 formulas that featured variations in the concentration of HPMC, including 1.5%, 2.5%, and 3.5%. Physical quality testing included the organoleptic test, homogeneity test, pH test, viscosity test, adhesion test, spreadability test, and real-time test. The data was analyzed using the SPSS system with one-way ANOVA and Paired T-Test. The results showed that variations in the concentration of HPMC as a gelling agent could affect the physical quality of gel preparations including viscosity, spreadability, adhesiveness, and pH, but did not affect organoleptics and homogeneity. The higher the concentration of HPMC applied, the viscosity, adhesion, and pH would also increase. However, the spreadability of the gel became low because it was inversely proportional to viscosity. The results yielded a good formula in formula 1 (HPMC 1.5%) and formula 2 (HPMC 2.5%).
