Abstract

NOVIA FITRI YANTI, 2025, UJI AKTIVITAS TONIKUM EKSTRAK SELEDRI (Apium graveolens L.) PADA MENCIT JANTAN (Mus musculus) METODE ROTAROD, KARYA TULIS ILMIAH, PROGRAM STUDI D-III FARMASI, FAKULTAS FARMASI, UNIVERSITAS SETIA BUDI SURAKARTA. Dibimbing oleh apt.Ismi Puspitasari, S.Farm., M. Farm. Seledri (Apium graveolens L.) merupakan tanaman yang mengandung senyawa flavonoid yang secara empiris dipercaya sebagai penambah stamina. Penelitian ini bertujuan mengetahui efek tonikum dan menentukan dosis efektif ekstrak etanol seledri yang dapat memberikan efek tonikum pada mencit jantan (Mus musculus). Uji tonikum digunakan untuk mengurangi kelelahan penambah stamina untk memenuhi kebutuhan sosial dan eknomi masyarakat dengan bertambahanya stamina tubuh. Efek tonikum merangsang dan memperkuat semua sistem organ dan merangsang perbaikan sel saraf pusat sehingga termasuk dalam kelas psikostimulant. Daun seledri mengandung senyawa flavonoid, saponin, tannin dan alkaloid. Penelitian ini dilakukan mengunakan metode rotarod dengan menghitung selisih waktu lelah mencit sebelum diberikan perlakuan (T0) dan setelah perlakuan (T1) . mencit sebanyak 25 ekor dibagi menjadi 5 kelompok dimana tiap kelompok terdiri dari 5 mencit. Kelompok I ( kontrol normal ) yaitu Na-CMC 0,5%, kelompok II (control positif) yaitu kafein 100mg/kgBB, kelompok III yaitu EES dosis 200mg/kgBB, kelompok IV yaitu EES dosis 400 mg/kgBB, kelompok V yaitu EES dosis 800mg/kgBB. Pada penelitian ini ekstrak etanol seledri memiliki efek tonikum terhadap mencit jantan(Mus musculus) yang diukur dengan metode rotarod dengan ditandai peningkatan waktu lelah yaitu waktu yang dibutuhkan mencit untuk tetap berada di atas alat rotarod. Hasil penelitian menunjukan bahwa ekstrak etanol seledri (Apium graveolens L.) mempunyai efek tonikum terhadap mencit jantan (Mus musculus) dengan dosis konsentrasi ekstrak 400mg/kgBB. Farm. Celery (Apium graveolens L.) is a plant containing flavonoid compounds, empirically believed to enhance stamina. This study aimed to determine the tonic effect and determine the effective dose of celery ethanol extract that could provide a tonic effect in male mice (Mus musculus). The tonic test is used to reduce fatigue and increase stamina to meet social and economic needs by increasing stamina. The tonic effect stimulates and strengthens all organ systems and stimulates the repair of central nervous system cells, thus categorizing it as a psychostimulant. Celery leaves contain flavonoids, saponins, tannins, and alkaloids. This study used the rotarod method, calculating the difference in fatigue time between mice before treatment (T0) and after treatment (T1). Twenty-five mice were divided into five groups of five. Group I (normal control) was given 0.5% Na-CMC; group II (positive control) was given 100 mg/kgBB of caffeine; group III was given 200 mg/kgBB of EES; group IV was given 400 mg/kgBB of EES; and group V was given 800 mg/kgBB of EES. In this study, celery ethanol extract had a tonic effect on male mice (Mus musculus) as measured by the rotarod method, indicated by an increase in fatigue time, the time required for the mice to remain on the rotarod apparatus. The results showed that celery (Apium graveolens L.) ethanol extract had a tonic effect on male mice (Mus musculus) at a dose of 400 mg/kgBB.
