Abstract

Diabetes mellitus type 2 was induced by high fat diet and fructose. The insulin resistance and hyperinsulinemia compensatory can be measured by insulin resistance test. Swietenia macrophylla King is a traditional plant can be used to treat diabetes mellitus and the main active compound of this plant is flavonoids dan triterpenoids. The objective of this study is to evaluate the effect of extract and fractions Swietenia macrophylla King seeds of high fat diet and fructose induced Wistar Rats. The research was conducted by using completely randomized posttest only control group design. Test animals used were divided into 10 test groups. Group 1: normal control; group 2: CMC Na 1%; group 3: metformin 45 mg/kg BW; group 4: ethanol extract 150 mg/kg BW; group 5: n-Heksan fraction 15 mg/kg BW; group 6: n-Heksan fraction 30 mg/kg BW; group 7: ethyl acetate fraction 15 mg/kg BW; group 8: ethyl acetate fraction 30 mg/kg BW; group 9: water fraction 15 mg/kg BW; group 10: water fraction 15 mg/kg BW. Animal induced insulin resistance by administration of High Fat Diet (HFD) and fructose. Data was analysed using One Way Anova (p < 0,05) continued by Dunnet T3 test. The results showed that extract and fractions Swietenia macrophylla King seeds had activity to decreased level of glucosa blood and increased GLUT 4 proteins in the most effective dose n-Heksan fraction Swietenia macrophylla King seeds 30 mg/ kg bw mice in which there was increased level of glucosa blood as 59,84% and and increased GLUT 4 proteins as 227,33. Result of statistical analysis (p<0.05) showed there was linear relationship between amount of increase in GLUT 4 protein with decrease percent in blood glucose levels.
