Abstract

In the pathological state such as diabetes mellitus, increased oxidative stress will cause decrease in endogenous antioxidant activity in the body so that the body do not able detoxify free radical and prevent cell damage. One source of natural antioxidant as antidiabetic is Moringa oleifera leave. The purpose of this study was determine antihyperglycemic activity and antioxidant activity of SOD and GPx enzyme and decrease MDA level of Moringa oleifera leave ethanol extract and fractions of Moringa oleifera leave ethanol extract. This study was an laboratory experimental research using a post-test only group design. The subject of this study were 50 male Wistar rats which conditioned type 2 diabetes by streptozotozin-nicotinamid induction. Rats were randomly divided into 10 groups, and each group consisted of 5 rats. Doses of ethanol extract fractions of Moringa leaves were used the fraction of n-hexane 32 mg/kg of rats body weight and 64 mg/kg rats body weight, the ethyl acetate fraction of 7.16 mg/kg rats body weight and 14 mg/kg rats body weight, the water fraction of 61 mg / kg rats body weight and 122 mg / kg rats body weight. The test results were showed that (1) the ethanol extract and fractions of ethanol extract of moringa leaves have antihyperglycemicactivity to the streptozotosin-nicotinamide-induced rats; (2) The ethanol extract and fractions of ethanol extract of moringa leaves can increase the activity of antioxidant enzymes of SOD, GPx and lower MDA levels; and (3) The ethanol extract and fractions of ethanol extract of moringa leaves increased the diameter of islet of langerhans and increase the population of the islet cells insulin production islet of langerhans.
