Abstract

Black cumin seed (Nigella sativa L.) is a plant that has property as traditional medicine. Chemical content of black cumin seed is saponnins, alkaloids, tannins, flavonoids and terpenoids. This study was conducted to determine the antibacterial activity of fraction of n-Hexane, ethyl acetate and water from 96% ethanol extract of black cumin seed (Nigella sativa L.) to Pseudomonas aeruginosa ATCC 27853. Extraction of black cumin seed is done by maseration metod using ethanol 96% solvent, then fractionated using n-Hexane, ethyl acetate and water solvent. The results of extraction and fractionation were tested for antibacterial activity of Pseudomonas aeruginosa ATCC 27853 using diffusion and dilution method. The diffusion method for knowing minimum inhibitory concentration and the most active fraction of Pseudomonas aeruginosa ATCC 27853 with concentration 50%, 25% and 12,5%. Dilution method is for knowing minimum bactericidal concentration of active fraction with concentration dilution series; 50%; 25; 12,5%; 6,25%; 3,125%; 1,562%; 0,781% and 0,390%. ANOVA oneway is used for statistical analysis to determine whether there is significant difference between the test preparation. The results howed that ethyl acetate fraction has the most active inhibition power compared n-Hexane and water fraction that 18,3 mm concentration to Pseudomonas aeruginosa ATCC 27853. Dilution test results showed antibacterial activity with MBC 12,5%. Keyword : antibacterial, fraction, black cumin seed (Nigella sativa L.), Pseudomonas aeruginosa.
