Abstract

Metoclopramide is an antiemetic drug that also acts as a dopamine antagonist, similar to procainamide as an antiemetic that works centrally in the area of Chemoreceptor Trigger Zone and peripheral by decreasing the sensitivity of the visceral nerves that deliver the afferent impulses from the gastrointestinal tract to the vomiting center. This study aims to determine the effect of Starch 1500® superdisintegrant variation and crosprovidone on the physical quality of ODT metoclopramide. The study was conducted using direct-compression method with three superdisintradate variation formulas, namely Starch 1500®, crosprovidone and combination of Starch 1500® and crosprovidone in different formulas. The resulting tablets are then tested for physical quality which includes: uniformity of weight, uniformity of size, hardness, fragility, uniformity of content, wetting time, crushed time, and sense response. Then analyzed using one way ANOVA statistic and SNK test at 95% confidence level. The results showed that the use of superdisintegrant Starch 1500® and crosprovidone affect to the physical quality of tablets. The excessive Starch 1500® concentration makes the tablets more fragile and disintegrate faster but has poor flow properties, while crospovideone is a superdisintegrant that has good flow properties so that combination of both materials will result in a qualified superdisintegrant.
