Abstract

Verapamil HCl is a calcium channel antagonists drug used for the treatment of cardiovascular disorders. Verapamil HCl has a bioavailability of about 20% with a half-life of 4 hours and low solubility at high pH, making it suitable for gastroretentive preparations. Gastroretentive dosage form one of them with floating system. This study aims to determine the effect of effervescent component concentration variation on the physical quality of floating verapamil tablet HCl and to know the concentration of citric acid and sodium bicarbonate which can produce the best physical quality of floating verapamil tablet HCl. Making floating verapamil tablet HCl using wet granulation method with variation of concentration citric acid and sodium bicarbonate 0%: 100%, 50%: 50%, and 25%: 75%. Testing of granules includes drying drift, flow time and still angle. Tests on tablets include tablet weight uniformity, tablet hardness, tablet fragility, floating lag time and floating time. The resulting data were analyzed statistically one-way anova with 95% confidence level. The results showed that the effect of citric acid can decrease hardness and speed up floating lag time and sodium bicarbonate can increase hardness and speed up floating lag time. Formula III with 25% citric acid concentration and 75% sodium bicarbonate can produce the best physical quality of floating verapamil tablet HCl with fast floating lag time compared to Formula I and Formula II.
