Abstract
Ketoprophen is a drug which has good permeability, low solubility in water and unpleasent taste. To improve its solubility and its taste through inclusion complex by β-cyclodextrin. In the developing to increase the release of ketoprophen is make it into the fast disintegrating tablets. The speed of drug release is influenced by the speed of the preparation to disintegrate. Combination sodium starch glycolate and crospovidone combine the action of swelling and wicking so as to speed up the disintegrating the tablets. The aim of this research to obtain the optimum value of the combination of both in order to obtain FDT wih optimum evaluation parameters according to the requirements. The study was made 8 run FDT with direct compression method with variation of concentration sodium starch glycolate 2–8% and crospovidone 2–5%. Evaluation performed at each run include flow velocity test, uniformity of dosage, hardness, friability, disintegrating time, wetting time and dissolution. The data were analyzed using Simplex Lattice Design with Design Expert version 7 application so as to obtain the optimum FDT formula. The optimum formula of the predicted result was verified by one sample t-test at 95%. The result showed inclusion complex 3.76 times increased solubility and 100% improved ketoprophen taste. Characteristic with FTIR showed the loss of the carboxylic and cabonil ketone groups in ketoprohen and with the DSC showed that endothermic peak did not appear in the inclusion complex, it indicated there has been an inclusion complex. An increase in proportion sodium starch glycolate increased hardness, disintegrating time and wetting time and decreased friability of FDT. The concentration sodium starch glycolate 2% and crospovidone 5% produced FDT with the optimum physical evaluation and drug released.
