Abstract
Bitter melon fruit and turmeric are plant that these plants are widely used as vegetable or spice in. This plant is known to be efficacious to treat diabetes mellitus. This study aimal to determine the effect of a combination of ethyl acetate fraction of bitter melon fruit and turmeric in increasing the levels of SOD and MDA lowering the GLUT-2 expression in rat liver insulin resistance. This study was conducted using a completely randomized direction. The animals were divided into 8 groups that each group consisted of 5 rat: Group I as normal control rat were given equivalent amounts of Na-CMC 1%, group II as a negative control were given feed HFD, lard and fructose without treatment, III as a positive control group were given metformin 9 mg / 200 g BW group IV ethyl acetate fraction of turmeric 10mg / 200g BB, group V bitter melon fruit fraction of ethyl acetate 0.4 mg / 200 g B, group VI-VIII were each given ethyl acetate fraction bitter melon fruit: turmeric (0.8 mg: 5 mg; 0.4 mg: 10 mg and 0.2 mg: 20 mg). All groups were given feed-fructose HFD for 45 days except the normal group that was given the standard feed. Administration of consecutive the fraction was carried out for 15 days. The results showed that the ethyl acetate fraction of bitter melon fruit and turmeric (dose combination III) and a single dose of turmeric could increase the levels of SOD and MDA enzyme in the liver of insulin resistance rat. The ethyl acetate fraction of bitter melon fruit and turmeric (dose combination III) and a single dose of bitter melon fruit succeded to increase the expression of GLUT-2 in hepatocytes cells of insulin resistance rat. In quantitative terms the occurrence of a single dose of bitter melon fruit ethyl acetate fraction (0.4 mg / kg) and fraction dose combination III (bitter melon fruit + turmeric 0.2: 20mg / Kg BW) which has a density of GLUT-2 protein that is comparable to controls positive is 28.677% and 29.474%.
