Abstract

Infection is a kind of disease that is mostly suffered in Indonesia. One of the cause of this infection is the E. coli bacteria. Most of the E. Coli can be found in the human’s large intestine as a normal flora. The purple leaf (Graptophyllum pictum (L.) can be used as an antibacterial. This research is being done in purpose to find out the activity of this antibacterial from the n-hexane, ethyl acetate and water’s fractions from the purple leaf’s ethanol extract towards the E. coli ATCC 25922. This study uses the maceration method using 96% ethanol and then been fractionated with the n-hexane, ethyl acetate, and water. The test method for the antibacterial activity that is used is the diffusion and dilution. The concentration of the ethyl acetate, n-hexane, and water of the purple leaf’s ethanol extract which is used each of diffusion method is 50%, 25% and 12,5%. The dilution is done by using a fraction of the most active of the diffusion. The test for the antibacterial activity using diffusion method the extract and the fractions of the purple leaf have the antibacterial activity. The ethyl acetate is the most active fraction. The diameter of the inhibition zone on the ethyl acetate fraction in the 50% concentration is 23,3 mm; 24 mm and 23 mm. The test for the antibacterial activity with the dilution method shows that the CMB is 12.5%. Key Words : Graptophyllum pictum (L.) Griff., Fraction’s, Escherichia coli, diffusion, dilution.
