Abstract
Tuberculosis is a infectious disease caused by Mycrobacterium tuberculosis bacteria, which can attacking various organs, especially lungs. Isoniazid is used to treat tuberculosis, most tuberculosis patients can be healed with isoniazid. The mechanism of action isoniazid inhibiting the synthesis micolat acid cell walls via an oxygen-dependent pathway. This study aims to determine the comparison of isoniazid levels in vitro to in vivo levels. This study uses healthy blood plasma given isoniazid levels 300 µg/mL in vitro and blood plasma of tuberculosis patients taking continuous phase antituberculosis drugs. Analysis of plasma levels using a C-18 HPLC column amd mobile phase potassium dihydrogen phosphate and acetonitrile 99:1. The data were analyzed by t-Test. The result showed that the ratio of isoniazid content in vitro to in vivo levels obtained in vitro levels is 8,525 µg/mL and in vivo levels is 5,091 µg/mL. Keywords : isoniazid, in vitro, in vivo, HPLC
