Abstract
Fisetin is flavonoid compound belonging to flavonol category useful as antioxidant agent. Fisetin belongs to the 2nd-grade BCS with low solubility of 10%, high permeability, and small water-solubility (0.002 mg/ml). Low solubility requires fisetin to be developed in the form of topical preparation, etosome, to improve drug penetration into skin. This research aimed to find out the effect of ethanol’s varying concentrations on fisetin etosome’s quality viewed from particle size, absorption efficiency and stability. Fisetin etosome was prepared using sonication-cool method. Cool method is a heating method at 300C to be shrunk later using sonication. This research employed fisetin 10 mg, phosphatidilcholine 75 mg, and ethanol in individual formulas of 25%, 30%, 35%, 40%. Etosome produced was characterized for its particle size, morphology, absorption efficiency and stability. Fisetin could be prepared using etosome carrier system in varying ethanol concentrations. The higher the ethanol concentration, the smaller was the particle size, the larger was the absorption efficiency and the more guaranteed was the stability. Particle sizes of all formulas were 271, 235, 402, and 196 nm, respectively. The largest absorption efficiency occurred in formula 2 and 4, 93.460% and 93.250%, respectively. Stability test showed that only formula 4 did not indicate precipitation. Keywords: fisetin, etosome, sonication-cool method, phosphatidilcholine.
