Abstract
Doxorubicin is one of the chemotherapy agent much used in the therapy for breast cancer. Therapy with doxorubicin have constraints that meant is important to combined with natural product like a Hydnophytum formicarum to reduce a toxic effect of an agent chemotherapy. The aims of this research is to know the activity of the Hydnophytum formicarum of cytotoxic as singular or combination with doxorubicin against proliferation lymphocytes, vero and mcf-7. The process of extracting the Hydnophytum formicarum should be conducted with the methods maceration by using a solvent ethanol and continued fractionate with chromatography liquid vacuum with a solvent ethyl acetate and ethanol with various comparison concentration. The cytotoxicity and the proliferation of lymphocytes activity uses the MTT assay. The form of absorbance converted into percent living cells. Percent cell viability is used to calculate the IC50 and then analyzed using ANOVA statistical analysis. Identification of compounds Hydnophytum formicarum performed by thin layer chromatography. The results showed that the ethyl acetate fraction of ethanol extract of Hydnophytum formicarum in combination with doxorubicin have a cytotoxic effect on MCF-7 cells and not toxic to vero cells. The combination of Hydnophytum formicarum and doxorubicin significantly different from control cells, the IC50 value of ethyl acetate fraction of 591.69 mg / mL and ½ IC50 0.395 nM doxorubicin. The analysis shows that the fraction of the study have immunostimulatory activity of the lymphocyte cell proliferation at a concentration of 73.96 mg / mL were significantly different compared with the positive control and the control cells. TLC results showed that the ethyl acetate fraction of ethanol extract of ant nests (Hydnophytum formicarum) contains flavonoids and phenolic.
