Abstract
Direct compression is the choosen method in the manufacturing of tablets. The process of making a few more steps and the stability of the active substance become the main reason to selected this method. All the advantages of direct compression method, this method has difficulties such as fluidity, compactibility and dissolution abilities. Not all physical properties of excipients have to be compressed. Development excipients with one co-processing excipients expected to overcome these limitations. This study aimed to develop co-processing excipients combine od Avicel PH 101 and PVP as filler binder in the manufacturing tablets by direct compression. Co-processed excipients are made using three formulas with ratio of 100% Avicel : 0% PVP; 50% Avicel PH 101 : 50% PVP and 0% Avicel PH 101 : 100% PVP. Material co-processed excipients tested physical quality as in granules. Further this material was used as filler binder of manufacturing tablet by direct compression which used Asetosal as a model of the active substance. Asetosal tablets tested physical quality. The parameters of the physical quality test material co-processed excipients and tablets optimized using the program Design Expert 8,0,6. Optimum formula of this study were obtained on a comparison 95% Avicel PH 101 : 5% PVP. The comparison optimum formula was get coprocessed excipients that fulfill the requirements fluidity and compactibility for direct compression. However, other requirements such as dissolution testing, the use of combination of Avicel PH 101 and PVP which inhibits of Asetosal dissolution rate. This is due of the hygroscopic nature of PVP. PVP often used as polimer in sustained-release tablet as to control the drug release. Keywords : co-processed excipients, Avicel PH 101, PVP, direct compression
