Abstract

Brotowali (Tinospora crispa L. Miers) is one of medicinal plants used to treat diabetes mellitus. Obesity and insulin deficiency is a risk factor of diabetes mellitus. This study aims to determine the effect of a decrease in blood glucose levels and the most effective dose infusion of stem brotowali comparison with the combination of metformin on blood glucose levels decrease in obese mice with alloxan induction. Test animals were divided into 7 groups. One group was given a normal diet and six groups were given a high-fat diet for 3 weeks and then induced with alloxan. Test preparation was administered for 14 days, blood glucose levels were checked on day 4, 9 and 14 after administration of the test preparation. Group I as a normal control, group II as negative controls given distilled water, group III was given metformin as a positive control (2,147 mg/33 g bw), group IV was given a single dose of brotowali stem infusion (92,45 mg/33 g bw), group V, VI and VII was given a combination of brotowali stem infusion and metformin 69,34 mg : 0,54 mg/33 g bw; 46,23 mg : 1,07 mg/33 g bw and 23,11 mg : 1,61 mg/33 g bw. Statistical data obtained by one way ANOVA (p <0.05). The results showed that the combination of metformin and infusion of stem brotowali able to provide comparable hypoglycemic effect with the use of metformin or a single infusion of stem brotowali. The combination of metformin and infusion of stem brotowali 75%:25% were able to provide a hypoglycemic effect and milder side effects. Brotowali stem infusion can be used as a complementary therapy oral antidiabetic drug metformin in patients with type 2 diabetes mellitus.
