Abstract

Amylase is a hydrolytic enzyme that breaks the bonds of glycosides in starch into smaller molecules such as glucose, maltose, and dextrin. Amylase produced from amylolytic bacteria is generally an extracellular enzyme. Amylase activity can be influenced by temperature and pH. Amylase in the pharmaceutical field is used as a biochemical marker of early diagnosis of acute pancreatitis and treatment of indigestion. The aim of this study was to isolate, test the activity and characterize amylase produced by bacteria. Amylase is obtained by fermenting each bacterium into the test medium for three days, then centrifuging at a speed of 5000 rpm for 10 minutes. Supernatant (crude amylase extract) obtained was tested for amylase activity in starch media, and iodine was added to the media to determine the clear zone formed. The next stage is characterization to test the resistance of amylase of each bacterium to temperature variations of 30, 40, 50, 60, and 70oC, as well as variations in pH 4, 5, 6, 7, and 8. The results showed that the five bacteria could produce amylase enzymes. The greatest amylase activity was derived from amylase Escherichia coli ATCC 25922 with a clear zone diameter of 19,79 mm. The characteristic test showed the results of Escherichia coli ATCC 25922 having the greatest amylase activity at 30oC and pH 8, while the amylase Pseudomonas aeruginosa ATCC 27853, Salmonella typhi ATCC 13311, Serratia marcescens ATCC 8100, Staphylococcus aureus ATCC 25923 had the highest activity at 50oC and pH 8.
