Abstract
Breast cancer ranks second after cardiovascular disease in developing countries. One plant that has the potential to be developed as an anticancer drug is white tumeric (Curcuma zedoaria). This study aimed to determine cytotoxic activity, selectivity index and protein number 53 in T47D cell culture. This research includes extraction of white tumeric using remaseration method with ethanol 96% solvent cytotoxic activity test of white methanolic extract was carried out by the MTT method [3- (4,5-dimethylthiazol-2 il) 2, 5 diphenyltetrazolium bromide) with series concentration (500 µg/ml;250 µg / ml;125 µg/ml;62.5 µg/ml;31.5 µg/ml) in T47D cells and calculated the value of concentration 50 IC50 inhibitors using linear regression. Cytotoxic selectivity is known by the selectivity index equation, namely the ratio of IC50 vero cells versus IC50 cells T47D. The amount of protein 53 is known by painting using an immunocytochemical method with an indirect method using primary and secondary antibodies. Test results of cytotoxic activity of ethanol extract of white rhizome using MTT method showed moderate cytotoxic activity in T47D cell culture with an IC50 value of 107 µg / ml. The selectivity index is 4,21. The amount of protein 53 uses an immunocytochemical method with an indirect method many brown cell cells express mutated 53 proteins.
