Abstract
Fisetin is a flavonoid compound potentially serving as antioxidant. The development of phycetine is conducted widely particularly in topical preparation. Fisetin is classified into 2nd grade- BCS (Biopharmaceutical Classification System) with good permeability and solubility of low. This can be overcome with making nano phytosome to improve drug penetration into skin. This research aimed to find out whether or not fisetin nanophytosome can be prepared using thin-layer hydration-sonicatin method, the effect of phosphatidilcholine’s varying concentrations on the quality of fisetin nano-phytosome, the profile of fisetin characterization after it is converted into nano-phytosome, and its stability during storage process. Fisetin nano-phytosome was prepared using thin layer hydrationsonication method, containing components the fisetin, phosphatidylcholine and cholesterol content ratio of 1:1:0.2; 1:2:0.2; 1:3:0.2; 1:4:0.2 and 1:5:0.2. Nanophytosome formula undertook physical characteristic test including particle size, penetration efficiency, stability, and antioxidant, then the data obtained was compared with literature. The result of the fisetin nano phytosome formula undertook physical characteristic test including particle size produced average particle sizes of 8811; 231.50; 152.67; 147.20; and 136.87 nm in F1, F2, F3, F4 and F5, respectively. The efficiency of penetration was measured from the third, fourth, and fifth formulas because it has qualified particle size range with the % penetration efficiencies of 83.72%; 87.29% and 88.42% in third, fourth, and fifth formulas, respectively. The antioxidant fisetin nano phytosome antioxidant test has a strong potential with IC50 of 68,257 ppm. Stability test was conducted only on the fifth formula but shows unstable results.
