Abstract

Ascorbyl palmitate has a high antioxidant level and is the derivation of vitamin C with low solubility and stability and cannot penetrate well into the stratum corneum, so that nanophytosome is developed to improve drug penetration into the skin. This research aimed to prepare nanophytosome using thin layer hidration-sonication method and to find out the effect of varying concentration of phosphatidilcholine on the quality of ascorbyl palmitate nanophytosome and its characterization viewed from its particle size, penetration efficiency, and stability aspects. Ascorbyl palmitate nanophytosome was prepared using thin-layer hydration-sonication method. Five (5) formulas were developed in this research with the following components: 10 mg ascorbyl palmitate, 1.83 mg cholesterol, and varying concentrations of phosphatidilcholine of 18.34 mg, 36.68 mg, 55.02 mg, 73,37 mg, and 91.71 mg. The characterization of nanophytosome included particle size, penetration efficiency, and stability and then the data obtained was compared with literature. Nanophytosome could be prepared from ascorbyl palmitate using thin layer hydration-sonication method. The characterization of ascorbyl palmitate nanophytosome produced average particle sizes of 96, 116, 97, 93, and 113 nm in F1, F2, F3, F4 and F5, respectively, with the largest penetration efficiency of 85.25% found in formulas 4. Stability test was conducted on formula 4 only with its particle size of 1105 nm and zeta potential -13,040 mV, meaning that ascorbyl palmitate nanophytosome was not stable during storage system.
