Abstract

Red Betel is one of the plants that has been widely studied because it empirically has the properties of curing various diseases and is scientifically proven as an anticancer. This study aimed to determine the cytotoxic activity, selectivity, and immunocytochemistry of red betel leaf ethanol extract against liver cancer cell culture (HepG2). This study included extraction of red betel leaf using maceration method with 70% ethanol. Cytotoxic test of red betel leaf ethanol extract was carried out by MTT method using concentration series (2000; 1000; 500; 250; 125; 62,5; 31,2; 15,6) µg / ml and IC50 values were calculated using linear regression. The selectivity index to determine cytotoxic selectivity is known by calculating between the IC50 values of vero cells compared with the IC50 value of HepG2 cells. Immunocytochemical reactions to determine the expression of Caspase-3 protein in the cancer cell apoptosis pathway were carried out using a series of IC50 concentrations (2000 µg / ml), ½IC50 (1000 µg / ml), and ¼IC50 (500 µg / ml) and calculated percentage of cell expression in each -one concentration. Cytotoxic test results of red betel leaf ethanol extract showed less potent activity on HepG2 cells with IC50 value 443,250 µg / ml, but had high cytotoxic selectivity with selectivity index value 3,323. In the immunocytochemistry test the red betel leaf ethanol extract was able to increase the expression of caspase-3 to induce cancer cell apoptosis.
