Abstract
Cervical cancer is one type of disease whose cases are increasing in the world. The prevalence of this disease increased in Indonesia from 1.4 per 1000 population in 2013 to 1.79 per 1000 population in 2018. Purslane is a type of garden weed that can thrive in subtropical to tropical environments. Purslane is traditionally used as an ornamental plant in Indonesia, and is still rarely used as a treatment. Purslane is traditionally used as an anti-inflammatory, analgesic, hepatoprotective and antioxidant. Purslane contains flavonoid compounds, especially campferol, apigenin, miricetin, quercetin, luteolin, carotene and alkaloids. The purpose of this study was to determine the cytotoxic effect of 70 % ethanol extract and the fraction of purslane herb (Portulaca oleracea L.) on cervical cancer cells (HeLa) and to determine the best fraction of purslane herb (Portulaca oleracea L.) which had a cytotoxic effect on cervical cancer cells (HeLa). This study included the extraction of purslane herbs with 70% ethanol using the maceration method and then liquid-liquid fractionation using n-hexane and ethyl acetate as solvents. Cytotoxic test using the MTT assay method against HeLa cancer cells and the IC50 value is known by looking for linear regression between the concentration log and cell viability. The results showed that the extract, ethyl acetate fraction, and n-hexane fraction had activity against cervical cancer cells (HeLa) with an average IC50 value of 105.6679, respectively; 46.0880; 53.1108 g/ml. Purslane extract has moderately active cytotoxic activity with IC50 values of 100-500 g/ml, while the ethyl acetate fraction and n-hexane fraction have active cytotoxic activity with IC50 values of 10-100 g/ml. Keywords: purslane herb (Portulaca oleracea L.), cytotoxic, cervical cancer, cells (HeLa), MTT assay
