Abstract
Vitamin E also known as α-tocopherol, is an antioxidant present in olive oil. Because α-tocopherol is non-polar and fat soluble, it necessitates the use of a non-polar solvent to extract α-tocopherol molecules. To acquire the levels of α- tocopherol in Extra Virgin Olive Oil (EVOO), this study needed to create a solvent capable of producing the highest levels of α-tocopherol in a substance. The specific gravity and refractive index of olive oil samples were determined before HPLC analysis. The maximum wavelength was determined using a UV-VIS analyzer and different comparisons were carried out using HPLC derived from the values of tR, N, HETP, and TF at flow rates of 0.8 mL/minute, 1 mL/minute, and 1.5 mL/minute. The technique is then validated, which includes linearity, LOD, and other factors. The results showed that the optimum conditions for HPLC were a maximum wavelength of 286 nm, a flow rate of 1 mL/minute and an injection volume of 10 μL. The level of-tocopherol obtained in Extra Virgin Olive Oil (EVOO) olive oil is 0.145 ppm with a correlation coefficient (r) of 0.998, percent recovery value of 99.77 percent, and percent KV of 1.53 percent.
