Abstract
Type 2 diabetes mellitus has several protein targets as therapeutic targets. Sambiloto is a plant that is believed to have efficacy in overcoming diabetes mellitus. Traditional medicines from plants generally form complex interactions with many targets. network pharmacology is a method that can be used to reveal the complex interactions of a pathophysiological pathway. This study aims to determine the compounds in herbs that are able to interact with target protein, and to determine the pharmacology profile. This research uses cytoscape software to build network pharmacology. Sample in this study was the chemical content of the bitter herbs obtained from the knapsack database and the target protein was obtained from the SuperTarget database. Gene names are validated with database from uniprot and then look for protein-protein interactions using a String database. Interactions on String database with combined score of 0.9 and above were collected as data. All data that has been obtained are tabulated. Tabulated data is then imported into Cytoscape to build tissue pharmacology. The visualization result shows that caffeic acid and beta-sitosterol involved in DPP4 network as DPP4-inhibitor lead to inhibition of incretin degradation, which causes the release of glucagon to be inhibited, thereby increasing insulin secretion, chlorogenic acid involved in INSR network as PTP1B inhibitor.
