Abstract
Depression is an emotional disorder in the long term caused by decreased levels of neurotransmitters in the brain. Papaya leaves (Carica papaya L.) contain alkaloids, flavonoids, saponins, and tannins which are used as antidepressants. The purpose of this study was to determine the antidepressant activity and effective dosage of the ethanol extract of papaya leaves (Carica papaya L.). In this study, using papaya leaf ethanol extract with antidepressant test and forced swimming test method by observing immobility time in mice. A total of 25 male mice were divided into 5 test groups. The first group was given CMC Na 0.5% as a negative control. The second group was given amitriptyline at a dose of 0.065 mg/20 g BW of mice as a positive control. The papaya leaf extract group (Carica papaya L.) was prepared using the maceration method and 96% ethanol solvent with 3 variations in dosage (150; 250; and 350 mg/kgBW). Each test group had previously been induced depression for 7 days by means of swimming. After that, the mice were treated until day 14. Immobility time observations in mice were observed on days 1, 7, and 14. The results of this study indicate that papaya leaf extract has antidepressant activity as seen from the immobility time using the forced swimming test method. The most effective dose to significantly reduce immobility time in mice is a dose of 350 mg/kg BW. Keywords : Antidepressants, papaya leaf extract, immobility time, forced swimming test
