Abstract

Isoniazid is a nicotinic acid derivate and act as the most effecticative tuberculosis agent againts micobacterium tuberculosis and a bacterisid spectrum for a rapit growth bacillus. The best time to administer isoniazid is 1-2 hours before meals. Previous research has shown that dietary intake can reduce the concentration of isoniazid in plasma and elevate Tmax isoniazid. This study aims to determine the effect of drug eating rules on isoniazid levels if consumed before and after meals This study test was using blood plasma tuberculosis patients consisting of 3 patients taking the drug before meals and 3 patients taking the drug after eating. In this research, examination levels of isoniazid in blood plasma tuberculosis patients by High Performance Liquid Chromatography (HPLC) method using a reversed phase C18 ODS colomn and an isocratic solvent programme, mobile phase was the mixture of potassium dihydrogen phosphate pH 6.2-acetonitrile (99: 1) with a flow rate 1 ml/ min and measurement were done at a wavelength of 230 nm. The data were analyzed by t-test, used paired samples ttest. The peak area of isoniazid at different concentrations with correlation coefficient of 0.993 and from the calculation results obtained regression equation y = 34740,43x + 34582,27. From the results of this study, it was found that levels isoniazid before meals 15,45 µg/mL and after meals 5,09 µg/mL it shows a differences in isoniazid levels if used before and after meals.
