Abstract

The teki grass (Cyperus rotundus L.) is a versatile weed plant, which is widely used in traditional medicine. Puzzles (Cyperus rotundus L.) contain flavonoids, alkaloids, saponins, and tannins which have antibacterial activity. The purpose of this study was to determine the antibacterial activity of ethanol extract, n-hexane fraction, ethyl acetate, and water fraction of puzzles on Pseudomonas aeruginosa ATCC 27853. Extraction was carried out by maceration using 96% ethanol, then fractionated using nhexane, ethyl acetate and water solvents. Antibacterial activity testing uses diffusion and dilution methods. Diffusion method to obtain the inhibition zone diameter from extracts and fractions. The diameter of the inhibition zone was analyzed using ANOVA data. The dilution method was carried out in the series of the most active fraction dilution and ethanol extract in the test tube to obtain the MIC value, after which inoculation was carried out to obtain the KBM value. The results showed that ethanol extract, n-hexane, ethyl acetate, and water fractions had antibacterial activity. The 10% concentration of ethanol extract, n-hexane, ethyl acetate, and water fractions gave inhibitory activity of 17 mm, 18.5 mm, 17.76 mm, 16.26 mm respectively. The 10% n-hexane fraction is the most active fraction. The KBM value of the ethanol and n-hexane extraction fraction is 10%. Keywords: Teki grass, Pseudomonas aeruginosa, diffusion, dilution
