Abstract

The compound curcumin is a compound of polyphenols wich is used as antibacterial. The curcumin analogues compounds research with the adding of the bromo group has the same activity or better as antibacterials. The purpose of this study was to determine the activity of curcumin analogues compounds 2,6-bis- ((5´-bromo-2´-furanyl)methylene)siklohexanone (compound D), 2,5-bis-((5´- bromo-2´-furanyl)methylene)siklopentanone (compound E), and 1,5-bis-(5´- bromo-2´-furanyl)-1,4-pentadien-3-on (compound F) to the bacteria Proteus mirabilis ATCC 10975 and Proteus mirabilis resistant siprofloksasin as an antibacteria and its mechanism of action. Antibacterial activity of the compound was determined by dilution method with acetone with a concentration of 10.000 ppm to determine Minimum Bactericidal Concentration (MBC) to the bacteria Proteus mirabilis ATCC 10975 and Proteus mirabilis resistant siprofloksasin. The most effective compound was made concentration series 0x, 1x and 2x MBC for analysis of leakage of cells ( nuleic acids and proteins) with the spectrophotometry UV-VIS, metal ions (K+ dan Na+) with Atomic Absoption Spectroscopy (AAS). The results showed that the compound F to the Proteus mirabilis bacteria resistant siprofloksasin is the most effective compound as an antibacterial from the third test compounds with the concentration MBC 19,531 ppm. Compound F can damage the cell walls and cell membrane permeability change marked by the reléase of nucleic acids, proteins, ions K+ dan Na+ of Proteus mirabilis resistant siprofloksasin.
