Abstract

Use of theophylline combination with cimetidin for asthma with GERD can increase blood levels of theophylline. The research was aimed to observe cimetidin and extra doses orally cimetidin which can provide significant changes the pharmacokinetics profiles of theophylline in Wistar rats. The study followed a completely randomized design used male Wistar rats were divided into 3 groups, the control and 2 treatment groups, each consisting of three test animals. The control group were given single theophylline 11.7 mg/kgBW. Treated groups II were given theophylline 11.7 mg/kgBW with cimetidin 36 mg/kgBW and groups III were given theophylline 11.7 mg/kgBW with cimetidin 18 mg/kgBW. Blood samples (0.2 ml) taken serially for 12 hours withdrawn at plexus retroorbitalis via the eye for HPLC with bufer acetate:asetonitril (93:7) as a mobile phase analysis of unchanged theophylline in blood. The concentration of theophylline was determined based on standard curve. The concentration-time data determines theophylline pharmacokinetics. The results indicated that cimetidin not affected the pharmacokinetics profiles of theophylline and doses 36 mg/kgBW not change the pharmacokinetics profiles of theophylline in Wistar rats.
