Abstract

Propranolol HCl is a non-selective β-adrenergic receptor-blocking agent, it is used as antihypertensive, antiarrythmic and antianginal. Propranolol has half-life about 2 hours and absorbed on gastric thus benefit formulated in floating drug delivery system to increase bioavaibility and reduce frequency of drug adminstration. This study was purposed to optimize and evaluate the influence of citric acid and sodium bicarnonate in physical properties, floatation behaviour, drug release of floating propranolol tablet and determine influence of pH of medium in floating lag time and swelling index. This study used factorial design method to optimize floating propranolol tablet using citric acid and sodium bicarbonate as independent variables. Hardness, floating lag time, swelling indeks, Q60, Q180 dan Q300 used as critical parameter to determine optimum area using software Design Expert® verse 7.1.5. Enhancement of citric acid and sodium bicarbonate level reduced floating lag time and increased drug release on 60, 180 and 300 minutes. Enhancement of citric acid reduced hardness of tablet whereas sodium bicarbonate increased swelling index on medium pH 1,2. Based on superimposed contour plot, the optimum area was citric acid 11,16 mg and sodium bicarbonate 72 mg. Keywords : citric acid, sodium bicarbonate, propranolol floating tablet
