Abstract
Theophylline is a bronchodilator used for asthma and to cope with chronic obstructive pulmonary disease. Almost 90% of theophylline experience biotransformation in the liver by the enzyme cytochrome P-450 (CYP) and only 10% excreted through the kidneys in unchanged form. Enzymes involved are CYP 1A2, 2D6, 3A3/4 and 2E1. Fuji apple is often consumed by the public. Fuji apple contain flavonoid called quercetin. Quercetin has high antioxidant activity and may inhibit the enzyme cytochrome P-450 sub CYP1A2, CYP2E1, CYP3A4. This study was aimed to determine the pharmacokinetic profile of theophylline when administered simultaneously with fuji apple juice. This study using rabbit test animals as 9 animals divided into 3 groups. The first group, control group theophylline dose of 7 mg/KgBW, group II theophylline 7 mg/KgBW and fuji apple juice 4 g/KgBW, and group III theophylline dose of 7 mg/KgBW with double fuji apple juice 8 g/KgBW. Analysis of plasma levels using HPLC column C-18 and mobile phase acetate buffer and acetonitrile 93:7. Snippets of blood was taken at minutes of 15, 30, 45, 60, 120, 180, 240, 300, 360, 420, 480, and 540. The data obtained were analyzed by ANOVA. The results obtained, fuji apple juice single dose could increase Tmax 27.00% and double dose 19.37%, increase AUC single-dose 43.11% and double dose 29.22%, and decrease CLT single dose 31.48%, double dose 19.37%. This parameter indicates that quercetin found in apples fuji could inhibit the metabolism of theophylline when administered simultaneously.
