Abstract

Multiple sclerosis (MS) is a complex autoimmune disease that damages the central nervous system (CNS) because of the attack of innate and adaptive immune system that causes demyelination, loss of oligodendrocyte, axonal degeneration and inflammation. The content of steroid contained in takokak fruit (Solanum torvum Swartz), are expected to provide immunosuppressive effect in reducing the number of microglia in MS. This research aimed to determine the effect of n-hexane fraction of takokak fruit against histopathological and number of spinal cord microglia and the most effective dose of MS rat. A total of 35 rats were divided into 7 groups, each group consisting of 5 rats. Group 1 as the normal control group was not treated, group 2 as the negative control were given Na CMC 0.5% and group 3 as the positive control were given methyl prednisolone 45 mg/kg BW. Groups 4, 5, 6 and 7 are given n-hexane fraction with successive doses; 70 mg/kgBW, 140 mg/kgBW, 280 mg/kgBW and 560 mg/kgBW. On day 0, groups of 2-7 induced with 0.2 ml encephalitogen sc and 0.5 ml Pentabio® vaccines sc. On 3rd day, groups of 2-7 induced again with 0.5 ml Pentabio® vaccine sc. Groups 2-7 were given the test preparation from day to 14-30. Each day, the rats were weighed and observed the neurologic symptoms. On day 31, the rats were sacrificed and spinal cord histopathology test. Data were analyzed by one way ANOVA using SPSS. The results showed that the fraction of takokak fruit (Solanum torvum, Swartz.), affect the histopathological picture and the number of spinal cord microglia of rat model of MS with the effective dose of 280mg/kg BW.
