Abstract

Vitamin C which also called ascorbic acid has important role in metabolism process, the characteristics of this vitamin are easy to be damaged by oxidization, heat and alkali. Vitamin C is available on long beans vegetable. The purpose of this research is to know whether raw long beans as well as boiled long beans has vitamin C substance and can be established using UV-Vis Spectrophotometry. Sample used in this research was long beans vegetable (Vigna sinensis L) with 3 treatments, i.e. raw, boiled at 80˚C and boiled at 100˚C. The method used to detect vitamin C amount started from the qualitative analysis of starting reaction and antioxidant test thin layer-chromatographically using silica gel GF254 movement elused using chloroform methanol (7:3) , continued by giving DPPH solution reactant and followed by the quantitative analysis using UV-Vis spectrophotometry method. Analysis method which used is standard curve method to generate linear line equation, y = a + b x d, by using 6 variations of standard solution concentration of vitamin C. The rest of sample absorption which obtained is calculated by linear regression equation, so group C can be converted to amount calculation. The result of research indicated that there is difference in amount of vitamin C in each samples. The percentages of vitamin C amount are 0,93% in raw long beans sample, 0,035% in sample which boiled at 80˚C, and 0,0049% in sample which boiled at 100˚C.
