Abstract

Diabetes mellitus (DM) is a metabolic disease characterized by increased blood glucose levels due to insufficiency of insulin function. Phaleria macrocarpa [Scheff] Boerl potential as antihyperglycemic. This study aims to determine antihyperglycemic activity and effect on the expression of GLUT-2 in cells of the pancreas and liver cells of the ethanol extract of mahkota dewa meat fruit ethanol extract in Streptozotosin Nicotinamid induced rats. The quantity of sample as much as 25 strain Wistar male white rats were randomly divided into 5 groups. Group 1: normal control, group 2: negative control, group 3: positive control, group of 4: EBMD 100 mg/kgbw and group 5: EBMD 200 mg/kgbw. The treatment was given orally for 14 days. Parameters measured were blood glucose levels, the expression of GLUT-2 in cells of the pancreas and liver. Data percentage decrease in blood glucose levels were analyzed by one way ANOVA statistical test and Tukey HSD. The results showed that EBMD dose of 100 mg/kgbw and 200 mg/kgbw for 14 days can lowed blood glucose levels significantly with significant value 0,000 respectively (p <0.05) compared to the negative control. EBMD for 14 days qualitatively able to increase the expression of GLUT-2 in pancreatic cells with those in the normal control, negative control, positive control, EBMD 100 mg/kg bw, and EBMD 200 mg/kgbw respectively to 100%; 32.31%; 67.67%; 59.56%; 66.09%. EBMD qualitatively able to reduce the expression of GLUT-2 in liver cells with those in the normal control, negative control, positive control, EBMD 100 mg / kg and EBMD 200 mg / kg respectively to 13.21%; 100%; 52.05%; 89.32%; 79.48%.
