Abstract

Diabetes Mellitus (DM) type II occurs in individu with insulin resistance or pancreatic β cells damage that was not capable to respond glucose to produce insulin. Insulin is an important factor of glucose transport. Glucose transporter (GLUT)-2 on pancreatic β cells membrane is responsible for glucose transport that will stimulated insulin secretion. The medical plants with potential to increase secretions of insulin and antioxidant can be used to therapy of DM. The purpose of this research was to determine the antyhyperglicemic activity and the effect to GLUT-2 protein expression in pancreatic β cells and hepatocyte STZ-NA induced rats. This research was used five groups of Wistar male rats, the first group as normal control. The second group as negative control (STZ-NA induced). The third group as a positive control (glibenclamide + STZ-NA). The fourth and fifth groups as test groups fenugreek ethanol-extract and was given 100 mg/kg BW and 200 mg/kg BW dosage for 14 days. Conducted measurement of blood glucose levels, observation of GLUT-2 expression of pancreatic β cells and hepatocyte rats by immunohistochemistry. The results showed that the test group extract of fenugreek seed was given 200 mg/kg BW, decrease blood glucose levels that was equivalent to the positive control. Incresed levels of GLUT-2 density in pancreatic β cells and decreased levels of GLUT-2 density in hepatocyte cells significantly occured at 200 mg/kg BW compared with 100 mg/kg BW dosage. Thus the ethanolic extract of fenugreek seed 200 mg/kg BW dosage was capable to decrease blood glucose levels, increase of GLUT-2 density in pancreatic β cells and decrease of GLUT-2 density in hepatocyte cells in STZ-NA induced rats that the effectiveness was comparable to positive control.
