Abstract
Suruhan Herba (Peperomia pellucida (L.) Kunth) is a plant that can be used in the treatment of type II diabetes mellitus. The study aimed to determine the antidiabetic activity of the ethanol extract of the Suruhan herba and determine the activity on GLUT-4 protein translocation muscular tissue in insulin resistance rats. The research was conducted using test animals Wistar rats (Rattus norvegicus) were used to divided into 6 groups of tests. Group 1: normal control; Group 2: negative control (Na-CMC); Group 3: positive control (Metfomin 45 mg / kg); groups 4, 5 and 6 is a variation of the dose groups: ethanol extract dose groups of 100, 200 and 300 mg / kg. Animal testing of diabetes mellitus is made by feeding a high-fat diet is the composition of feed pellets (80%), lard (15%), and a quail egg yolk (5%). Pellet feed given to the animals every day test ad libitium while giving lard and quail eggs orally administered daily in test animals. The whole test animals given high fat diet treatment and fructose every day for 45 days. Normal control animals not given a high-fat diet treatment and fructose. Fructose is given once per day by oral doses of 180mg / 100g BB. Fasting glucose assay reagent kit. Observations translocation of GLUT-4 protein in rat muscle tissue using immunohistochemical techniques. Antidiabetic activity using three parameters, namely: (1) testing of blood glucose levels preprandial and postprandial, (2) HE staining (3) observation GLUT-4 protein translocation to the thigh muscular tissue. Results showed that the ethanol extract of suruhan herba dose of 300 mg / kg BW within 15 days of the hypoglycemic power has given percentage of 18.03 ± 2.605 and the dose has shown an increase in activity of the translocation of GLUT-4 protein in muscle tissue by 83.22% against rats were conditioned to experience insulin resistance, type II diabetes as a result of feeding a high-fat diet.
