Abstract

Red betel (Piper crocatum) and Phaleria (Phaleria macrocarpa) are medicine plants that can be used to overcome type II diabetes mellitus type II. This study aimed to determine the antidiabetic activity of combination of ethanolic extract of red betel leaf and ethanolic extract of phaleria, and determine the effect on GLUT-2 expression in pancreatic β cells and decrease of GLUT2 expression in liver tissue in streptozotosin-nicotinamide-induced rat. The study was conducted using Wistar rat (Rattus norvegicus) divided into 5 groups. Group 1: normal control; Group 2: negative control (Na-CMC); Group 3: positive control (glibenclamide 5 mg/kg BW); groups 4 and 5 were combination group: ethanolic extract of dose groups 50:100 and 100:200 mg/kg BW. The diabetes mellitus was induced with streptozotosin-induced 50 mg/kg BW and nicotinamide 110 mg/kg BW for group 1-5 intraperitoneally. Determination of fasting glucose levels used GOD-PAP Kit . Observation of GLUT-2 expression in pancreatic β cells and liver tissue used immunohistochemical technique. The antidiabetic activity used three parameters, i.e.: (1) test fasting blood glucose levels, (2) observation of GLUT-2 protein expression in pancreatic β cells (3) observation of GLUT-2 protein expression in liver tissue. The results showed that red betel extract dose of 50 mg/kg BW and phaleria extract 100 mg/kg BW (combination I) within 14 days exhibited percentage of hypoglycemic effect by 24.32 ± 2.07 % anas well as activity to increase GLUT-2 protein expression in pancreatic β cells as 43.16% and to decrease of GLUT-2 expression in liver tissue as 71.07% to rat conditioned DM type II result of streptozotosin-nicotinamide-induced.
