Abstract

Acetylcholine (ACh) is the neurotransmitter at neuromuscular junctions, visceral ganglia motoric system, and various parts of the central nervous system. Acetylcholine hydrolysis by the enzyme acetylcholinesterase (AChE). Acetylcholine plays a role in the regulation of cognitive functions, especially in encoding of new memories. Noni is widely available in Indonesia and had been proven to have the ability of AChE enzyme inhibition. This study aimed to determine the ability of nonpolar-semipolar fractions of ethanol extract of noni fruti to inhibits the enzyme acetylcholinesterase activity and chemical compounds in the active fraction of the ethanol extract of noni fruit. Separation of chemical compounds was done through a two-stage extraction using maceration method and fractionated using vacuum liquid chromathography. AChE enzyme inhibition testing conducted by the Ellman’s method (colorimetric method). Identification of chemical compounds was conducted by thin layer chromathography and liquid chromathography-mass spectrometry. IC50 value of the fraction I and fraction II of noni fruit showed respectively at 927,893 and 1976,60 μg / ml. IC50 value of the ethanol extract and fraction III of noni fruit can not be determined (> 2000 ug / ml). Thin layer chromathography skrining showed fraction I contained terpenoid and phenolic groups, fraction II countained terpenoid, steroid, phenolic, coumarin groups and fraction III countained terpenoid, steroid, phenolic, flavanoid, anthraquinon groups. Fraction I is active fraction and expected contained 3,3-bisdemethylpinoresinol and pinoresinol.
