Abstract

Meloxicam is a selective of cyclooxygenase-2 enzyme that has been used as rheumatoid arthritis treatment. Meloxicam is poorly water-soluble drug, furthermore inclusion complex in the β-cyclodextrin can be used to improve its solubility and mask the bitterness taste of the meloxicam. Formulation of fast disintegrating tablet (FDT) meloxicam increase patient compliance. This study purposed to determine the influence of the combination of superdisintegrants Kollidone CL and Primojel in FDT meloxicam on the physical properties of tablets and drug release in β- cyclodextrin inclusion complex. Inclusion complexes was prepared by kneading method using 1:2 drug to β- cyclodextrin molar ratio and characterized using thermal analysis (difference scanning colorimetric; DSC), solubility and drug release. Direct compression method was applied in tablet preparation. Kollidone CL (A) and Primojel (B) has been used as independent variabels that consist of 5 formulations and determined on physical properties. The results were statistically analyzed using analysis of variance (ANOVA, p-value set 0,05). The DSC confirmed that meloxicam and β-cyclodextrin complex was formed due to disappearance of meloxicam melting point in complex. The drug release of inclusion complexes increased solubility and drug release than that of meloxicam. Kollidone® CL provided an influence of increasing the friability and decreasing the disintegration time, wetting time, meanwhile Primojel® increased wetting time and the disintegration time and reduced friability. Kollidone® CL and Primojel® with the same concentration FDT produce tablets with lower friability and shorten disintegration time.
