Abstract

Angkak (red yeast rice) was a rice obtained by fermentation using Monascus sp. Angkak contained flavonoid compound with potency of natural antioxidant, so it can be used as anti wrinkle. Angkak was made in form of extract, then in order to achieve an optimum effect use of angkak extract needed a good delivery systems in the form of nanoemulsion. And to easy of use then nanoemulsi formed in cream. Purpose of the research was to know antioxidant activity of angkak extract, a good nanoemulsion formulation and its cream preparation that was examined for penetration test and safety. Nanoemulsion was made by mixing the oil phase, surfactant and cosurfactant selected, red yeast rice extract and water phase, then rotated with ultra turrax speed of 1500 rpm for 30 minutes. Cream made into a type of oil in water (O / W). Cream was divided into three formulas, where in: formula 1 was negative control (KN), formula 2 was angkak extract cream (KEA), and formula 3 was nanoemulsion angkak extract cream (KNE). Antioxidant activity of angkak extract was examined by using DPPH method. Cream tested stability, safety test with primary irritation index value and ocular and penetration tested by Franz diffusion cells. The results of the research indicated that the angkak extract had potent antioxidant activity with IC50 value of 2.60 ppm. Nanoemulsion selected with a ratio of surfactant and cosurfactant, tween 80 and glycerin (1:3) with globule size of 152.267 nm, polydispersity index of 0.271 and zeta potential of -14 mV. KNE had higher penetration test than KEA. The cumulative amount of angkak extract penetrated for 2 hours of KNE of 2288.02± 1.43 ug / cm2 while the KEA for 766.26± 0.53 ug / cm2. Safety test showed KNE that the cream a little irritation with IIPR value of 1.67 and for IIO no irritation to the value of 0.
