Abstract
Syzygium cortex has been used as antidiabetic agent. Diabetes mellitus type II is due to resistant and function deficiency of insulin. This research aimed to determine the activity of ethyl acetate fraction of Syzigium cumini L. cortex toward reducing of blood glucose and enhancement of translocation of GLUT-4 on Wistar rat with insulin resistant. The animals were divided in to 7 groups, i.e. normal control (group 1), negative control using Na CMC 1% (group 2), positive control using metformin (group 3), ethyl acetate fraction 25, 50 and 100 mg/kg body weight (BW) (group 4, 5 and 6, respectively) and ethanol extract 150 mg/kg BW (group 7). The animals were administrated fructose and high fat diet (HFD) until insulin resistant condition was achieved. Blood glucose was measured after administration of HFD, 30 days and 50 days as pretreatment and post treatment was observed at 58 and 65 days. Insulin resistant was tested by fasting blood glucose and translocation of GLUT-4 protein in muscular tissue. The result were analyzed statistically using one way ANOVA confidence interval level of 95% (p = 0.05). The results showed that ethyl acetate fraction 100 mg/kg BW was the most optimum could reduce the blood glucose and increased the translocation of GLUT-4 protein. Decreasing of blood glucose associated with increasing of GLUT-4 expression statistically (p<0.05). Key word: Syzygium cumini L, insulin resistance, translocation, GLUT-4
