Abstract
Phyllanthus niruri L. has been used as a medicine to reduce inflammation. The study aims to determine antiinflammatory activity and safety of the gastrointestinal tract as well as the class of chemical compounds responsible for that activity. The research was conducted by completely randomized method. In the in vivo antiinflammatory test, animals were divided into eight groups: negative control; positive control diclofenac sodium 9 mg/kgBB; Phyllanthus niruri ethanol extract (PNEE) 200, 100, 50 mg/kgBB; Phyllanthus niruri fraction of nhexane (PNFH), ethyl acetate (PNFEA), water (PNFW) 100 mg/kgBB. Animals induced with 0.1 ml of carrageenan and edema volume was observed from the first hour up to six hour. The next day the animals were administered the sample orally 1x daily and observed the safety of the gastrointestinal tract on the 6th day, macroscopic and histopathologic test. In vitro antiinflammatory test using membrane stabilization methods induced hipotonic solution with concentrations 100, 200, and 400 ppm. Each test seen of the absorbance at Uv-vis spectrophotometry. Results of in vivo antiinflammatory activity showed that the PNFH indicate highest activicy was significant with diclofenac sodium (p<0.05). In vitro antiinflammatory test showed PNFEA concentration 400 ppm was able to stabilize the erythrocyte membrane. PNEE 200 mg/kgBB and PNFEA 100 mg/kgBB is relatively safe in the gastrointestinal (in vivo). Chemical compounds responsible for the antiinflammatory activity in PNEE were flavonoids, tannins, saponins, and steroids whereas in PNFEA were flavonoids and tannins.
