Abstract
One of plant used empirically to decrease blood glucose level is Leucaena (Leucaena leucocephala (Lmk) De Wit) seeds. The research purpose was to determine inhibitory activity of α-glucosidase enzyme of leucaena seeds ethanol extract by in vitro and in vivo as well as antidiabetic effect in mice which inducted alloxan from Leucaena seeds ethanol extract. The method used in this study was pseudosubstract, glucose, sucrose, amylum tolerance tests and alloxan induction method. α-glucoside will hydrolyze p-nitrophenyl-α-D-glucopiroside substract into p-nitrophenyl and glucose. The enzyme activity was measured based on p-nitrophenyl absorbance in yellow coloured. Mice animal tests were grouped into 6 groups. Group I (negative control) was given Na.CMC, group II (positive control) was given acarbose dose of 10 mg/kg and test group III - VI were given Leucaena ethanol extract with dose of 1 ; 0.5 ; 0.25 ; 0.125 g / kg. Data which obtained processed statistically with ANOVA test. The result shows that Leucaena seeds ethanol extract could inhibit activity of α-glycosidase enzyme by IC50 value as 396.52 ppm, whereas acarbose had IC50 as 116.68 ppm. Treatment using glucose, sucrose and amylum tolerance tests showed that in treatment with glucose, the decrease of blood glucose levels in all groups were not significantly difference (ρ<0.05). This because the acarbose and Leucaena extract work by inhibit α-glucosidase activity so that glucose as monosaccharide was not affected by its absorption. The experiment result using sucrose and amylum tolerance tests was showed decrease in blood glucose levels significantly (ρ>0.05), the most glucose level decrease in positive control group of acarbose and Leucaena ethanol extract dose of 1; 0,5 and 0,25 g/kg BW mice. Whereas negative control did not shows decrease in blood glucose levels significantly. This because acarbose and Leucaena extract works by inhibiting α-glucosidase enzyme so sucrose as disaccharide and amylum as polysaccharide were inhibited its absorption.
