Abstract
Theophylline has a relatively short half-life is 3-7 hours and the narrow therapeutic index of 10-15 µg / ml and 125 mg dose usage is. Theophylline in tablet dosage formulation sustained release theophylline concentrations are expected to result in a more uniform and the blood peak levels that do not fluctuate. Based on this, research on the manufacture of slow-release theophylline tablets using HPMC and Na-CMC matrix. Interaction between Na-CMC with HPMC very dominant in determining the dissolution of theophylline. This study is based on the method simplex lattice design (SLD) with two matrix components, namely: (100% HPMC), (50% HPMC: 50% Na-CMC), (100% Na-CMC). Flow time, violence and DE360 as a parameter optimization. Then calculated using the design expert to determine the optimum formula. The results showed that theophylline sustained-release preparations can be made, and kombinsi HPMC matrix and Na-CMC affected the physical quality and the release of theophylline tablets. The proportion that produces sustained release theophylline tablets with optimum physical quality is a formula containing 58,33% HPMC and 41,67% Na-CMC.
