Abstract

Pine bark (Pinus merkusii Jungh et de Vriese) contains flavonoid bioactive compound. Flavonoid content of pine bark is expected to improve body immunity against hepatitis B virus. This research aimed to find out the effect of pine bark (Pinus merkusii Jungh et de Vriese) and effective dose on the increase IgG and IgM titers in hepatitis B induced Balb /c mice. The extraction method used was remaceration method with 70% ethanol. About 30 mice (5 groups) were divided into two, each of which consisted of 15 mice (Label A), 15 mice (Label B). Group I, II, III was given doses of 0.26 mg/20 g mouse BW, 0.39 mg/20 g mouse BW, and 0.52 mg/20 g mouse BW. Group IV was given Stimuno at 0.13 mg/20 g mouse BW and group V was given aquaduct as negative control. All groups (Label A) were induced with hepatitis B vaccine (Engerix B) on the day-8 (after acclimatization) and (Label A + Label B) on the day-15 intraperitoneal. IgG and IgM serum was taken on day-22 and then tested for protein content in immunoglobulin using indirect ELISA technique. The result of research showed that pine bark extract had an effect of improving immunoglobulin G (IgG) and immunoglobulin M (IgM) titers in hepatitis B vaccine induced Balb/c mice. The effective dose of pine bark extract in improving IgG and IgM titers was 0.26 mg/20 g mouse BW. Keywords: Pine bark (Pinus merkusii Jungh et de Vriese), IgG, IgM, Hepatitis B.
