Abstract

Macrophage serves as APC (antigen presenting cell) antigen introducing process to T-cell. Pine bark (Pinus merkusii Jungh. & De Vriese) contains proantocyanidin compound presumably improving body immunity. This research aimed to find the effect of pine bark ethanol extract administration and the effect of increased dose of pine bark ethanol extract on macrophage phagocytosis in hepatitis B vaccine-induced Balb/c mice. The extraction method used for pine bark in this research was remaceration with ethanol 70% solvent. About 30 mice were divided into 5 groups, each of which consisted of 6 mice. Groups I, II, and III were given pine bark ethanol extract at doses 0,26 mg/20g mice BW, 0,39 mg/20g mice BW, and 0,52 mg/20g mice BW. Groups IV was given stimuno at dose 0,130 mg and V was given aqueduct as negative control. All groups were induced with hepatitis B vaccine (Engerix B) on the day-8 (after acclimatization) and -15 intraperitoneal. Mice were euthanized to undertake macrophage phagocytosis on the day-22. The result of research showed that the pine bark ethanol extract improved activity and capacity of macrophage phagocytosis in hepatitis B vaccine-induced Balb/c mice. The effective dose of Pine bark extract to improve macrophage phagocytosis activity and capacity was 0,39 mg/20 g mice BW.
